Is it possible to create one LLD rule with 2 or more Item Prototypes in it?

Yes, it's possible.

You can create as much prototypes as you need. No any special requirements for discovery. Just use it's macroses in prototypes.

Check for example Mounted filesystem discovery in built-in "Template OS Windows" template. There are 4 prototypes defined.
