How to eanble connection to data base instance

As part of our daily basis work, I have to enable monitoring set up for monitoring all the data bases inside a Server. where I have configured everything in that, but I am not able to connect to that data base instance from our Zabbix controller server, I can connect to that server from RDS and also, I could connect that server with telnet also, but when I try to connect data base it is throwing an error like Login timeout expired, I don't know where the problem is occurring. please someone help me in this.
